The aim of the study is to analyze the methods for determining the cytotoxicity of doxycycline on the wound healing process at the stage of preclinical studies and its potential for the development of antibacterial wound healing agents for local use. It was established that doxycycline at concentrations used in wound regeneration models is characterized by a favorable cytobiological profile and does not exhibit cytotoxic effects on skin and connective tissue cells. Candidates for doxycycline-based dosage forms, such as nanofibrillar membranes and hydrogels, exhibit high biocompatibility and contribute to the acceleration of reparative processes, as evidenced by an increase in the rate of wound closure (≈21–56%), increased re-epithelialization and decreased MMP expression.
